On April 1, 2026, RGL-236 for Injection, an mRNA drug jointly developed by Shanghai Regenelead Therapies Co., Ltd. and its wholly-owned subsidiary Guangzhou Revolead Therapies Co., Ltd. (collectively, "Regenelead"), received implied approval from the National Medical Products Administration (NMPA) of China for its Investigational New Drug (IND) application, clearing the way for clinical trials in osteoarthritis (OA). The IND clearance of RGL-236 marks another major breakthrough for Regenelead in the field of local LNP-mRNA delivery and regenerative repair.
01.Addressing a Clinical Pain Point: Osteoarthritis Treatment Urgently Needs a Breakthrough
Osteoarthritis is a common chronic degenerative joint disease characterized by progressive damage to articular cartilage, which can lead to pain, deformity, and functional impairment. According to the Global Burden of Disease study, approximately 595 million people worldwide had osteoarthritis in 2020, and this number is projected to reach about 1.1 billion by 2050. In China, the overall prevalence of primary osteoarthritis among people aged 40 and above has reached 46.3%, and among those aged 60 and above it exceeds 62%, with the disease burden continuing to rise.
Current clinical treatments mainly focus on symptom relief, including oral or topical non-steroidal anti-inflammatory drugs (NSAIDs) and intra-articular injections of corticosteroids and sodium hyaluronate. However, none of these drugs has been clearly shown to inhibit the deterioration of joint structure or delay disease progression; ultimately, patients often need to undergo joint replacement surgery. At present, no disease-modifying osteoarthritis drug (DMOAD) has been approved for marketing anywhere in the world, leaving a significant unmet clinical need in this field.
02.Innovative Mechanism: A Synergistic Breakthrough in Protein Optimization and mRNA Delivery
RGL-236 uses Regenelead's proprietary lipid nanoparticle (LNP) delivery system. Administered by intra-articular injection, it efficiently delivers mRNA encoding an optimized protein to target cells within the joint cavity, where the protein is expressed. This approach is designed to reduce immunogenicity, increase the depth of cartilage penetration, prolong the duration of drug action, and improve patient adherence. The expressed target protein is intended to promote the synthesis of type II collagen, thereby supporting the repair and regeneration of hyaline cartilage tissue and restoring the articular surface affected by osteoarthritis. RGL-236 has the potential to become the world's first approved DMOAD, addressing the growing treatment needs of the global market.
Dr. Cheng Liao | Founder and Chief Executive Officer of Regenelead
"Osteoarthritis affects the quality of life of hundreds of millions of patients worldwide, and existing treatments do little to halt disease progression. The clinical advancement of RGL-236 not only brings osteoarthritis patients closer to a new treatment option with regenerative and reparative potential, but also further validates our technology platform capabilities in LNP-mRNA delivery and local regenerative therapy, and is expected to provide a scalable R&D pathway for the treatment of other degenerative diseases or tissue injury disorders."
